Nicotinamide N-Methyltransferase from Human, Recombinant
Nicotinamide N-Methyltransferase (NNMT) methylates nicotinamide and other pyridine containing compounds. NNMT transfers a methyl group from S-adenosylmethionine (SAM) to nitrogen N1 of nicotinamide to produce 1-methylnicotinamide (MNA) and S-adenosylhomocysteine. MNA is a messenger molecule that increases neurite branching, serves as an anti-thrombotic, and has anti-inflammatory properties. The role of NNMT overexpression in cancers may be to alter epigenetic methylation patterns in two ways: by lowering the intracellular concentration of SAM, required by methyltransferase enzymes, and by depleting the available NAD+ by transforming nicotinamide to MNA. Sirtuins use NAD+ as a substrate to alter protein acetylation and ribosylation, including histone targets. Group: Enzymes. Synonyms: NNMT. Enzyme Commission Number: E.C. 2.1.1.1. Purity: > 95% by SDS-PAGE. Mole weight: 42.6 kDa. Stability: As supplied, 6 months from the QC date provided on the Certificate of Analysis, when stored properly. Storage: Store at -80°C. Also acts on benzimidazole, and the clostridial enzyme acts on adenine to form 7-α-D-ribosyladenine 5'-phosphate. nicotinate phosphoribosyltransferase
The enzyme, which is involved in pyridine nucleotide recycling, can form β-nicotinate D-ribonucleotide and diphosphate from nicotinate and 5-phospho-α-D-ribose 1-diphosphate (PRPP) in the absence of ATP. However, when ATP is available the enzyme is phosphorylated resulting in a much lower Km for nicotinate. The phospho-enzyme is hydrolysed during the transferase reaction, regenerating the low affinity form. The presence of ATP shifts the products/substrates equilibrium from 0.67 to 1100. Group: Enzymes. Synonyms: niacin ribonucleotidase; nicotinic acid mononucleotide glycohydrolase; nicotinic acid mononucleotide pyrophosphorylase; nicotinic acid phosphoribosyltransferase; nicotinate-nucleotide:diphosphate phospho-α-D-ribosyltransferase. nicotine blue oxidoreductase
The enzyme, characterized from the nicotine degrading bacterium Arthrobacter nicotinovorans, catalyses the reduction of "nicotine blue" to its hydroquinone form (the opposite direction from that shown). Nicotine blue is the name given to the compound formed by the autocatalytic condensation of two molecules of 2,3,6-trihydroxypyridine, an intermediate in the nicotine degradation pathway. The main role of the enzyme may be to prevent the intracellular formation of nicotine blue semiquinone radicals, which by redox cycling would lead to the formation of toxic reactive oxygen species. The enzyme possesses a slight preference for NADH over NADPH. Group: Enzymes. Synonyms: nboR (gene name). Enzyme Commission Number: EC 1.1.1.328. Storage: Store it at +4 ?C for short term. For long term storage, store it at -20 ?C?-80 ?C. Form: Liquid or lyophilized powder. EXWM-0241; nicotine blue oxidoreductase; EC 1.1.1.328; nboR (gene name). Cat No: EXWM-0241.
