Microsphere surfaces are separately cross-linked with specific antibodies. After incubation with the mixture to be tested, the two antibodies formed double anti-sandwich immune complexes with the antigen, thus keeping the spacing between the two microheres within 200 nm. By using the donor microsphere, the surrounding oxygen is converted into excited singlet oxygen and spreads to the acceptor microsphere within 200 nm, so as to generate a stable and efficient light signal at 615 nm when excited at 680 nm.
